【Patent Number: No. 7657500】Composition for Promoting Klotho Expression and Related Inventions

We obtained a new patent right on April 7, 2025.

Mesenchymal stem cells (MSCs) are expected to have various therapeutic effects, such as anti-inflammatory effects, and numerous studies have been conducted to treat intractable diseases. However, the detailed molecular mechanisms have not been fully understood.

Our company has identified that the conditioned medium/cell culture supernatant derived from human MSCs (MSC-CM), which contains the secretions of mesenchymal stem cells, has the effect of increasing the expression of the Klotho gene. The Klotho gene is thought to be closely related to the lifespan of the individual. Furthermore, we have identified TNFSF12/TWEAK (TNF super family 12/TNF-related weak inducer of apoptosis) as an effective component for increasing the expression of the Klotho gene in MSC-CM, and obtained a patent right based on this discovery. The various therapeutic effects of mesenchymal stem cells may be mediated through the increased expression of the Klotho gene, and this patent is considered to be a fundamental one.

About Klotho
It has been reported that mutations in the Klotho gene in mice show symptoms of premature aging, suggesting that it is an important gene that controls lifespan (1). There are also reports that the amount of Klotho protein in human serum is inversely correlated with age (2), suggesting that the decrease in Klotho protein with age may contribute to the progression of aging phenomena.

About the conditioned medium/cell culture supernatant
To culture cells, it is necessary to use a culture medium (liquid) for cell culture, in which cells proliferate. During the proliferation process, various secretory factors are released from the cells. The culture medium containing these secretions from the cells is called the conditioned medium/cell culture supernatant. Among various cells, mesenchymal stem cells that our company is researching are known to secrete various growth factors, cytokines, and exosomes with high therapeutic effects into the culture medium.
